| Analytical Conditions | 本品の粉末1gを共栓遠心沈殿管に入れ，水酸化カリウム試液5mL及びアセトニトリル5mLを加え，密栓して10分間振り混ぜた後，遠心分離し，上層を試料溶液とする．別に薄層クロマトグラフィー用アストラガロシドⅣ 1mgをメタノール2mLに溶かし，標準溶液とする．これらの液につき，薄層クロマトグラフィー〈2.03〉により試験を行う．試料溶液及び標準溶液10μLずつを薄層クロマトグラフィー用シリカゲルを用いて調製した薄層板にスポットする．次に酢酸エチル／メタノール／水混液(20：5：4)を展開溶媒として約10cm展開した後，薄層板を風乾する．これに希硫酸を均等に噴霧し，105℃で5分間加熱した後，紫外線(主波長365nm)を照射するとき，試料溶液から得た数個のスポットのうち1個のスポットは，標準溶液から得た黄褐色の蛍光を発するスポットと色調及びRf値が等しい． |
