| 分析条件 | 本品の粉末0.5gにメタノール10mLを加え，水浴上で5分間加温し，ろ過し，ろ液を試料溶液とする．別に薄層クロマトグラフィー用ベルゲニン1mgをメタノール1mLに溶かし，標準溶液とする．これらの液につき，薄層クロマトグラフィー〈2.03〉により試験を行う．試料溶液及び標準溶液10μLずつを薄層クロマトグラフィー用シリカゲル(蛍光剤入り)を用いて調製した薄層板にスポットする．次に酢酸エチル／エタノール(95)／水混液(100：17：13)を展開溶媒として約10cm展開した後，薄層板を風乾する．これに紫外線(主波長254nm)を照射するとき，試料溶液から得た数個のスポットのうちRf値0.5付近の1個のスポットは，標準溶液から得た暗青色のスポットと色調及びRf値が等しい． To 0.5g pulverized Mallotus Bark add 10mL of methanol, warm on a water bath for 5 minutes, filter, and use the filtrate as the sample solution. Separately,dissolve 1mg of bergenin for thin-layer chromatography in 1mL of methanol, and use this solution as the standard solution. Perform the test with these solutions as directed under Thin-layer Chromatography<2.03>. Spot 10μL each of the sample solution and standard solution on a plate of silica gel with fluorescent indicator for thin-layer chromatography.Develop the plate with a mixture of ethyl acetate,ethanol(95) and water(100:17:13)to a distance of about 10cm ,and air-dry the plate. Examine under ultraviolet light (main wavelengh:254nm):a principal spot with a dark blue color which appears at an Rf value of about 0.5 from the sample solution is the same as the spot from the standard solution in color and the Rf. |
